چکیده

The formation of the essential functional unit of the ovary, the primordial follicle, occurs during fetal life in humans. Factors regulating oogonial proliferation and interaction with somatic cells before primordial follicle formation are largely unknown. We have investigated the expression, localisation and functional effects of activin and its receptors in the human fetal ovary at 14–21 weeks gestation. Expression of mRNA for the activin hA and hB subunits and the activin receptors ActRIIA and ActRIIB was demonstrated by RT-PCR. Expression of hA mRNA increased 2-fold across the gestational range examined. Activin subunits and receptors were localised by immunohistochemistry. The hA subunit was expressed by oogonia, and the hB subunit and activin receptors were expressed by both oogonia and somatic cells. hA expression was increased in larger oogonia at later gestations, but was low in oocytes within newly formed primordial follicles. Treatment of ovary fragments with activin A in vitro increased both the number of oogonia present and oogonial proliferation, as detected by bromodeoxyuridine (BrdU) incorporation. These data indicate that activin may be involved in the autocrine and paracrine regulation of germ cell proliferation in the human ovary during the crucial period of development leading up to primordial follicle formation. Quantitative Analysis of GABAA Gamma Receptor Subunits in the Developing Embryonic Chick Forebrain

Objective(s) In this study we investigated the expression of GABAA receptor subunits during brain development. These receptors may change in the embryonic chick forebrain. Materials and Methodes The expression levels of four types of GABAA receptor gamma subunits (γ1, γ2, γ3 and γ4) were quantified in the embryonic chick forebrain at 32 hr, 3, 7, 14, and 20 days of incubation and day one aft...

P-234: Expression of Human Chorionic Gonadotropin (hCG) Hormone Using Chinese Hamster Ovary Cells

Background: Human chorionic gonadotropin (hCG) is a member of glycoprotein hormones family consist of two different non-covalently heterodimeric chains: alpha and beta subunits with 92 and 145 amino acids respectively. This hormone plays an important role in human reproduction and physiology especially for maintenance of the corpus luteum during the first months of pregnancy Materials and Metho...

P-152: Expression of Recombinant Human Luteinizing Hormone (hLH) in CHO Cells

Background: Human luteinizing hormone (hLH) stimulates steroid biosynthesis of the ovary, triggers ovulation and prepares androgen production of testicular Leydig cells. LH belongs to the family of glycoprotein hormones that are heterodimers consisting of a common α-subunit and specific β-subunit. hLH is necessary for clinical and infertility treatment. Recombinant DNA technology provides a use...

Expression pattern of messenger ribonucleic acid for the activin type II receptors and the inhibin/activin subunits during follicular development in broiler breeder hens.

The expression of mRNA for the activin type II receptors (ActRII and ActRIIB), follistatin, and the inhibin/activin subunits was investigated in the follicles of broiler breeder hens. Total RNA was isolated from individual granulosa and theca layers of the F1 through F5 follicles, a pool of the F6 and F7 follicles, the small yellow follicles, and from the combined granulosa and theca layers of ...
